Bioactive Bromotyrosine Derivatives from the Pacific Marine Sponge Suberea clavata (Pulitzer-Finali, 1982) ArchiMer
Moriou, Céline; Lacroix, Damien; Petek, Sylvain; El-demerdash, Amr; Trepos, Rozenn; Leu, Tinihauarii Mareva; Florean, Cristina; Diederich, Marc; Hellio, Claire; Debitus, Cecile; Al-mourabit, Ali.
Chemical investigation of the South-Pacific marine sponge Suberea clavata led to the isolation of eight new bromotyrosine metabolites named subereins 1–8 (2–9) along with twelve known co-isolated congeners. The detailed configuration determination of the first representative major compound of this family 11-epi-fistularin-3 (11R,17S) (1) is described. Their chemical characterization was achieved by HRMS and integrated 1D and 2D NMR (nuclear magnetic resonance) spectroscopic studies and extensive comparison with literature data. For the first time, a complete assignment of the absolute configurations for stereogenic centers C-11/17 of the known members (11R,17S) 11-epi-fistularin-3 (1) and 17-deoxyfistularin-3 (10) was determined by a combination of...

Circular RNA hsa_circ_0000523 regulates the proliferation and apoptosis of colorectal cancer cells as miRNA sponge BJMBR
Jin,Y.; Yu,L.L.; Zhang,B.; Liu,C.F.; Chen,Y..
Among the novel class of endogenous long non-coding RNAs, circular RNA (circRNA) is known as a key regulator in the development and progression of different cancers. Its function and mechanism in the tumorigenesis of colorectal cancer, however, has not been well studied. This study thus aimed to investigate potential regulation of colorectal cancer by circRNAs and the corresponding regulatory mechanism. We demonstrated that the expression of circRNA hsa_circ_0000523 (also known as circ_006229) was down-regulated in different colorectal cancer cell lines. It was also found that interference of hsa_circ_0000523 induced proliferation and suppressed apoptosis of colorectal cancer cells, the proliferation rate of which was reduced by the overexpression of...

Diversity and biological activities of the bacterial community associated with the marine sponge Phorbas tenacior (Porifera, Demospongiae) ArchiMer
Dupont, S.; Carre-mlouka, A.; Descarrega, F.; Ereskovsky, A.; Longeon, A.; Mouray, E.; Florent, I.; Bourguet-kondracki, M. L..
The diversity of the cultivable microbiota of the marine sponge Phorbas tenacior frequently found in the Mediterranean Sea was investigated, and its potential as a source of antimicrobial, antioxidant and antiplasmodial compounds was evaluated. The cultivable bacterial community was studied by isolation, cultivation and 16S rRNA gene sequencing. Twenty-three bacterial strains were isolated and identified in the Proteobacteria ( or classes) and Actinobacteria phyla. Furthermore, three different bacterial morphotypes localized extracellularly within the sponge tissues were revealed by microscopic observations. Bacterial strains were assigned to seven different genera, namely Vibrio, Photobacterium, Shewanella, Pseudomonas, Ruegeria, Pseudovibrio and...

Feasibility study of organic matter and Ammonium removal using loofa sponge as a supporting medium in an aerated submerged fixed-film reactor (ASFFR) Electron. J. Biotechnol.
Nabizadeh,Ramin; Naddafi,Kazem; Mesdaghinia,Alireza; Nafez,Amir Hosien.
Biofilm systems are efficient in the removal of organic matter and ammonium from wastewaters. In this study, loofa sponge, a natural product, was used as a supporting medium in an aerated submerged fixed-film reactor to evaluate its performance in removing organic matter and nitrogen from wastewater. Four pilot runs were performed with chemical oxygen demand (COD) concentrations of 100, 200, 300 and 400 mg l-1 to provide an organic loading rate of 0.6, 1.2, 1.8, and 2.4 kg m-3d-1 respectively. In these pilot runs, the influent ammonium nitrogen concentrations were justified to 5, 10, 15 and 20 mg l-1 as N to provide an influent nitrogen loading of 30, 60, 90 and 120 g m-3.d-1 respectively. Although soluble COD removal efficiency greater than 80 percent was...

Immunization by subcutaneous implants of polyester-polyurethane sponges coupled with antigen BJMBR
Lima,S.L.; Machado,C.B.; Pereira,M.A.C.; Cara,D.C.; Velarde,D.T.; Andrade,S.P.; Gontijo,C.M..
A new protocol is described for immunization of outbred Swiss mice. The procedure is based on subcutaneous implantation of antigen-coupled polyester-polyurethane sponges cut into disks of 10 mm in diameter vs 2 mm in thickness. Antigen coupling was performed by overnight incubation of the sponge with a solution of ovalbumin (Ova) (2 mg/ml) diluted in sodium carbonate buffer, pH 9.6. The amount of ovalbumin that was taken up by the sponge was between 71.4 to 82.5 µg. This was estimated by comparing the Ova absorbance at 280 nm in coating buffer solutions before and after incubation. To compare the efficiency of the proposed method, experimental groups immunized with the antigen in the presence of adjuvants (10 µg in Al(OH)3 or 100 µg in complete Freund's...

Le mercure, le cadmium et le plomb dans les eaux littorales libanaises: apports et suivi au moyen de bioindicateurs quantitatifs (éponges, bivalves et gastéropodes) ArchiMer
Nakhle, Khaled.
In 1975, the meeting for the protection of the Mediterranean Sea in Barcelona fixed several important objectives. One of them was the creation of National Observation Programs in order to evaluate the levels and tendencies of pollutants. Furthermore the MED-POL program has re-evaluated and developed these objectives. Lebanon has signed the Barcelona Convention and has been engaged in providing data about its coastal water quality. Unfortunately the Lebanese war, which also began in 1975, paralyzed the country for 15 years. Since 1990, successive governments in the country have fixed as an objective the reconstruction of the tourism sector. As a consequence, they became interested in the environmental problems. The studies carried out before or during the...

Lock, stock and two different barrels: comparing the genetic composition of morphotypes of the Indo-Pacific sponge Xestospongia testudinaria Naturalis
Swierts, T.; Peijnenburg, K.T.C.A.; Leeuw, C. de; Cleary, D.F.R.; Hörnlein, C.; Setiawan, E.; Wörheide, G.; Erpenbeck, D.; Voogd, N.J. de.
The giant barrel sponge Xestospongia testudinaria is an ecologically important species that is widely distributed across the Indo-Pacific. Little is known, however, about the precise biogeographic distribution and the amount of morphological and genetic variation in this species. Here we provide the first detailed, fine-scaled (<200 km2) study of the morphological and genetic composition of X. testudinaria around Lembeh Island, Indonesia. Two mitochondrial (CO1 and ATP6 genes) and one nuclear (ATP synthase β intron) DNA markers were used to assess genetic variation. We identified four distinct morphotypes of X. testudinaria around Lembeh Island. These morphotypes were genetically differentiated with both mitochondrial and nuclear markers. Our results...

Marine Natural Occurring 2,5-Diketopiperazines: Isolation, Synthesis and Optical Properties ArchiMer
Laville, Remi; Thanh Binh Nguyen,; Moriou, Celine; Petek, Sylvain; Debitus, Cecile; Al-mourabit, Ali.
Seven 2,5-diketopiperazines (DKPs) were isolated from the Fijian marine sponge Acanthella cavernosa. NMR and circular dichroism (CD) comparison with synthetic L-L DKPs allowed us to determine unambiguously the L-L absolute configuration of the natural DKPs. This work initiated the setting up of an optical properties database of natural DKPs, including specific rotation and CD.

Microbiological and functional evaluation of an alternative device (OB®) for estrous synchronization in ewes Ciência Rural
Martins,Leonardo Tondello; Santos Neto,Pedro Claudino dos; Gaudêncio Neto,Saul; Rauber,Lúcio Pereira; Bertolini,Marcelo; Vieira,Arnaldo Diniz; Mezzalira,Alceu.
The use of synthetic progestagens released by vaginal devices is an important tool to overcome the reproductive seasonality in sheep, but cost and/or subsequent vaginitis are limiting factors for their use. To identify economic, simple and innocuous alternative vaginal devices for estrous synchronization/induction protocols in sheep, this study aimed to evaluate the microbiological and functional viability of the human vaginal tampons (OB®) impregnated with medroxyprogesterone acetate (MAP) on reproductive performance of ewes. The study compared them with commercial vaginal inserts (CIDR®) and polyurethane sponges impregnated with MAP. In Experiment 1, the device loss rate, the degree of vaginitis during the device removal, the count and identification of...

Preliminary biological characterization of the Saldanha hydrothermal field at the Mid-Atlantic Ridge (36 degrees 34 ' N, 32 degrees 26 ' W, 2200 m) ArchiMer
Biscoito, M; Almeida, A; Segonzac, Michel.
A preliminary description of the biology of the Saldanha hydrothermal field in the Mid-Atlantic Ridge (36 degrees 34'N, 32 degrees 26'W, 2200 m) is given, based on direct observation, video images and samples. Venting occurs mainly in a diffuse way through sediment, but hydrothermal fluid discharges are visible through centimeter-long orifices, scattered on the sediment, over an area of approximately 400 m(2), at depths between 2,200 and 2,220 metres. Emitted fluids are transparent, with a temperature of 7-9 degrees C. An almost complete absence of macro- and megafauna around the venting orifices was found. The only vent invertebrate identified was the gastropod Phymorhynchus sp., with only two juvenile specimens caught. A careful examination of the images...

Quorum Sensing Inhibitory and Antifouling Activities of New Bromotyrosine Metabolites from the Polynesian Sponge Pseudoceratina n. sp. ArchiMer
Tintillier, Florent; Moriou, Céline; Petek, Sylvain; Fauchon, Marilyne; Hellio, Claire; Saulnier, Denis; Ekins, Merrick; Hooper, John N. A.; Al-mourabit, Ali; Debitus, Cecile.
Four new brominated tyrosine metabolites, aplyzanzines C–F (1–4), were isolated from the French Polynesian sponge Pseudoceratina n. sp., along with the two known 2-aminoimidazolic derivatives, purealidin A (5) and 6, previously isolated, respectively, from the sponges Psammaplysilla purpurea and Verongula sp. Their structures were assigned based on the interpretation of their NMR and HRMS data. The compounds exhibited quorum sensing inhibition (QSi) and antifouling activities against several strains of bacteria and microalgae. To our knowledge, the QSi activity of this type of bromotyrosine metabolite is described here for the first time.

Soft Collagen-Gelatine Sponges by Convection Drying BABT
Meyer,Michael; Trommer,Kristin.
This study showed that thermally labile fibrillar collagen could be processed continuously in combination with gelatine as foaming additive by convection drying. The procedure led to stable sponges with similar structural and physical properties as found for freeze-dried collagen samples. The fibrillar collagen remained native, while gelatine acted as foaming additive. The absorbency of the sponges was improved by opening the surface with abrasives. A use as medical device with hemostyptic properties would be possible.

Sponge-Inspired Dibromohemibastadin Prevents and Disrupts Bacterial Biofilms without Toxicity ArchiMer
Le Norcy, Tiffany; Niemann, Hendrik; Proksch, Peter; Tait, Karen; Linossier, Isabelle; Rehel, Karine; Hellio, Claire; Fay, Fabienne.
Since the banning of several families of compounds in antifouling (AF) coatings, the search for environmentally friendly AF compounds has intensified. Natural sources of AF compounds have been identified in marine organisms and can be used to create analogues in laboratory. In a previous study, we identified that dibromohemibastadin-1 (DBHB) is a promising AF molecule, leading to the inhibition of the activity of phenoloxidase, an enzyme involved in the attachment of mussels to surfaces. This paper describes the activity of the DBHB on biofilm formation and its detachment and on bacterial adhesion and communication: quorum sensing. DBHB has an anti-biofilm activity without affecting adhesion of marine and terrestrial bacteria at a dose of 10 mu M....
